Any benefit? I hope not as I mowed it down and spread more clover seed in this plot

It has OK wildlife value. Ducks feed on it mostly. It is a hardy annual that could possibly out compete your clover.

What could I do to kill it, Spray with Roundup?

Best bet is a pre emergent like Oust XP. It is an annual so round up will take care of the current cohort but not the seed bank. Not sure what Oust will do to the clover.
